چکیده
Ester prodrugs are commonly used to increase the intestinal absorption of drugs with permeabilitylimited absorption. The high functional activity of este rases in blood and liver allows a rapid bio acti va tion of the prodrug after reaching the sys temic circu lation. However, the increased efficiency of prodrugs to pass the intestinal barrier may be decreased by its rapid esterase-mediated hydrolysis at the level of the intestinal mucosa, offsetting the diffusion of the lipo philic, intact esters across the epithelial monolayer. This phenomenon primarily occurs at the level of the mucosa of small intestine, due to its high esterase activity. Tenofovir disoproxil fumarate [(TENDF) is a bis-ester prodrug of the acyclic nucleoside phos pho nate tenofovir. The chemical name of TENDF is 9[(R)-2-[[bis[[(iso propoxy carbonyl) oxy]methoxy] phosphinyl] methoxy] propyl] adenine fumarate (1:1) (Figure 1). It has a molecular formula C19H30N5O10P JMB 2009; 28 (3) DOI: 10.2478/v10011-009-0013-z

Pharmacokinetic interactions between lersivirine and zidovudine, tenofovir disoproxil fumarate/emtricitabine and abacavir/lamivudine.
BACKGROUND To investigate pharmacokinetic interactions associated with coadministration of lersivirine with zidovudine, tenofovir disoproxil fumarate (DF)/emtricitabine (Truvada(®)) or abacavir/lamivudine (Epzicom(®)/Kivexa(®)). METHODS Three Phase I, open, crossover studies with two (studies 1 and 3) or three (study 2) treatment periods were conducted in healthy individuals. In study 1, indi...
متن کاملDevelopment of AgNPs-tragacanth Conjugated Gel as a Novel Green Membrane in Electro-membrane Extraction: Tenofovir Disoproxil Fumarate Assay in Human Plasma Samples
AgNPs-Tragacanth conjugate gel as an eco friendly and low cost membrane was applied for the effective electromembrane extraction (EME) of Tenofovir disproxil fumarate, an important acidic and high polar (log p=1.25) antiviral drug, followed by HPLC-UV determination. The effect of various parameters on the extraction efficiency including the presence of AgNPs, tragacanth gum concentration, gel t...

Objective: Development of an accurate, simple, precise and rapid method for estimating Tenofovir disoproxil fumarate, Lamivudine and Efavirenz simultaneously, in a combined tablet form. Methods: The method uses Reverse Phase High Performance Liquid Chromatography (RP-HPLC). Symmetry C18 (4.6 x 100 mm, 3.5 μm) column operated with a mixture of phosphate buffer of pH 4.0 with ortho phosphoric aci...
متن کاملDevelopment and validation of reversed-phase HPLC gradient method for the estimation of efavirenz in plasma
Efavirenz is an anti-viral agent of non-nucleoside reverse transcriptase inhibitor category used as a part of highly active retroviral therapy for the treatment of infections of human immune deficiency virus type-1. A simple, sensitive and rapid reversed-phase high performance liquid chromatographic gradient method was developed and validated for the determination of efavirenz in plasma. The me...
متن کاملA comparative pharmacokinetic and tolerability analysis of the novel orotic acid salt form of tenofovir disoproxil and the fumaric acid salt form in healthy subjects
A novel orotic acid salt form of tenofovir disoproxil (DA-2802) was developed and is expected to replace the fumaric acid salt form. The pharmacokinetic (PK) characteristics and tolerability profiles of DA-2802 were compared to those of tenofovir disoproxil fumarate (TDF, Viread®) in healthy subjects. A randomized, open-label, single-dose study was conducted in 36 healthy subjects using a two-t...
